Red Light Induced Change in the Saponin Content of Prolamellar Bodies in A vena sativa

چکیده
Etioplast, Protochlorophyllide, Prolamellar Body, Saponin, Red Light Exposure to red light of high intensities leads to saponin and prolamellar body degradation in etiolated Avena sativa seedlings. Exposure to red light of low intensities increases the saponin percentage found in prolamellar bodies without changing the overall saponin content. Proto chlorophyllide is assumed to act a decisive part in the phototransformation of the PLB as expressed by their saponin content. This is indicated by the action spectrum of this stimulation as well as by experiments with long-wave red (729 nm) and ALA/LA treatment. In weak light the pchlide phototransformation is decelerated-probably due to lack of energy. The PLB are inter preted to be modified by the increased pchlide concentration so that more saponin can be incorporated. In further experiments a barley mutant (albina f 17) lacking the capacity to esterify phytol as well as the capacity to degrade the PLB during the greening process was investigated. The results substantiate our hypothesis, as PLB transformation — indicated by PLB saponin concentration — is decelerated with increasing pchlide concentration in greening etioplasts.

Completely dissolved components of native prolamellar body/prim ary thylakoids PLB/PT were induced to reaggregate into tubular structures in vitro. Incubation of these pchlide con taining PLB-like tubules under red light with NADPH : pchlide oxidoreductase led to a specific association of the enzyme protein to the tubules. Photoconversion of the pigment initiates the dispersal of reaggregated ...

Developmental changes in fine structure were studied in plastids of etiolated bean leaves during the time required for the protochlorophyllide-chlorophyllide transformation and the following lag phase prior to chlorophyll accumulation. In agreement with some other workers, two distinct stages of change in the fine structure of proplastids were found to occur upon illumination during this period...

Etioplasts, Prolamellar Bodies, Saponins, Chromatography Prolamellar body tubules in etioplasts are composed mainly of two saponins. The determination of these saponins in leaves, etioplasts or subfractions of etioplasts is re commended to evaluate the concentration of these tubules in a given fraction. Using a densitometer after TLC-separation of the saponins measurements are carried out in a...

Prolamellar Bodies, Thylakoids, Chlorophylls Etioplast membranes were fractionated into enriched prothylakoid (PT) and prolamellar body fraction (PLB) by known procedures [2]. The photoconversion of Protochlide to Chlide was 77% in the PT fraction but only 65% in the PLB fraction with optimum NADPH concentrations. The subsequent esterification reaction proceeds in both fractions indicating that...
متن کاملRed-light stimulation of prolamellar body recrystallization and thylakoid formation in barley etioplasts.
The etioplasts of fully dark-grown barley leaves exhibit a relatively low frequency of crystalline prolamellar bodies (ca. 16-20%). Brief red-light treatment leads to rapid disruption of all prolamellar bodies followed by a slow reformation in the subsequent dark period. When several red-light treatments are given with intervening 3-h dark periods, a marked increase in the proportion of crystal...
